[en] The test set shows a container bowl, in which are several cylindrical containers are closely piroted in a carrier plate opposite to the container bowl. Each container pasesses a lower outlow. In each container a disc out of anti-body containing cellulose paper is arranged. The cellulose foil retaines the liquids used, is however permeable for therm when there is a depression generated in the container bowl. The porous structure at the cellulose paper guarantees the contact between the anti-bodies, the antigenes and the tracer material. (DG)

Die Testvorrichtung weist eine Aufnahmeschale auf, in der mehrere Zylinderbehaelter in einer Trageplatte dicht gegenueber der Aufnahmeschale aufgehaengt sind. Jeder Behaelter besitzt einen unteren Auslass. In jedem Behaelter ist eine Scheibe aus mit Anitkoerper versehenen Cellulosepapier angeordnet. Das Cellulosepapier haelt die verwendeten Fluessigkeiten zurueck, ist jedoch durchlaessig, wenn in der Aufnahmeschale ein Unterdruck erzeugt wird. [en] A double antibody sequential radioimmunoassay for human myelin basic protein (MBP) has been developed. The assay utilizes a rabbit antibody to human MBP and purified rabbit MBP as the radiolabelled antigen. This assay was used to analyze cerebrospinal fluid (CSF) from 22 patients with severe head injury, 61 other cases of various neurological disorders, and 106 normal controls. The results showed that closed head trauma caused moderate to severe elevations in CSF MBP, and elevated CSF MBP was detectable in several diseases which involve CNS myelin

[en] The dexametazone suppression test (DST) is a very valuable neuroendocrin assay as auxiliar method for diagnosing the depression. The serum cortisol levels of 20 patients of the Hospital de Salud Mental and of a control group were determined by radio-immunoassay technique. Normal concentration was defined as 5 micrograms per deciliter. Among depressed patients the average value was 9.8865 micro-grams per deciliter; having the test a 85% of positiveness. Among the control group the average value found was 1.167 micro-grams per deciliter, with a 20% of positiveness for the test. (author)

[en] Purification of human prolactin from pituitaries was carried out in our laboratory to obtain a pure reagent for use in RIA. The extraction and purification procedure was adapted from the method of Mc. Lean et al., and it involves the following steps: 1. Extraction of frozen pituitaries in buffers 0.14M phosphate/citrate pH 4.0 and 0.05M ammonium acetate pH 10.0. 2. Purification by hydrophobic interaction chromatography on Phenyl-Sepharose CL-4B in the presence of acetonitrile. 3. Purification by anion exchange chromatography on DEAE-Sepharose Cl-68. The purification method is considered effective for obtaining a hPrl of the purity needed for radioassay purposes, having the advantage of rapidity and relative simplicity. (author)

A purificacao de prolactina humana a partir de hipofises foi desenvolvida em nosso laboratorio com o objetivo de obtermos um reagente puro para uso em radioimunoensaio. O procedimento de extracao e purificacao foi adaptado do metodo de McLean et al., o qual envolve as seguintes etapas: 1. Extracao de hipofises congeladas em tampao fosfocitrato 0,14M, pH 4,0 e tampao acetato de amoneo 50 mM em pH 10,0. 2. Purificacao por cromatografia de interacao hidrofobica em Phenyl-Sepharose CL-4B na presenca de acetonitrila. 3. Purificacao por cromatografia de troca ionica em DEAE-Sepharose CL-6B. O metodo de purificacao foi considerado eficaz para obtencao de hPrl com a pureza necessaria para a preparacao de reagentes usados em radioimunoensaios, possuindo a vantagem de ser rapido e relativamente simples. [en] A radioimmunological method to determine total thyroxine in a serum sample is described. The method is as follows: trichloracetic acid and sodium hydroxide are mixed with the sample; radioactive thyroxine is added to the mixture, which is left to reach equilibrium then placed in contact with a resin sponge consisting of a polyurethane foam with intercommunicating cells containing a strongly basic anion exchange resin; the mixture and the resin sponge are incubated, the initial radioactivity of the mixture and resin sponge combination is measured with an appropriate detection system, then the resin sponge is removed from the mixture, washed and its residual radioactivity measured
