AbstractAbstract
[en] RNA replication of the pepper ringspot virus, its translocation and its association with mitochondria are studied. Some basic aspects of the research are first examined: actinomycin D (AMD) effects on parts of the nucleolus, nucleus and cytoplasm of healthy - and infected cells; comparative study between the circle method and the planimetry method to determine the cell areas; determination of the proportion between the silver grain densities of nucleulus, nucleus and cytoplasm of the cells treated with AMD; determination of the HD (Half-Distance) for the working conditions. Use of the mathemathical model proposed by NADLER gives basic information with respect to the translocation and association of the virus with the mitochondria in the host cells: in the mitochondria associated system the silver grains covering the two components are predominantly constituted by the RNA of the radioactive virus (78%); the time necessary for the RNA synthesis, the virus maturity and its translocation to the mitochondria, (checked by U-5-3H treatment) can be shorter than 5 hours. Estuda-se a replicacao do RNA do virus do anel do pimentao (VAP), sua translocacao e associacao com mitocondrios. Examinam-se inicialmente pontos fundamentais para o desenvolvimento da pesquisa: efeito da actinomicina D (AMD) sobre as areas do nucleolo, nucleo e citoplasma das celulas sadias e infectadas; estudo comparativo entre a determinacao das areas celulares pelo metodo dos circulos e planimetria; determinacao da proporcao existente entre as densidades de graos de prata do nucleolo e do nucleo e a densidade destes graos no citoplasma das celulas tratadas com AMD; obtencao do valor do HD (Half-Distance) para as condicoes de trabalho. Aplicacao do modelo matematico proposto por NADLER a translocacao e associacao VAP- mitocondrio da celula hospedeira, permite obtencao de duas informacoes basicas: no sistema VAP- mitocondrio associados, os graos de prata cobrindo os dois componentes sao predominantemente formados pelo RNA radioativos do VAP (78%) e, o tempo necessario para sintese do RNA, maturacao e translocacao do virus ate os mitocondrios coincide com o tratamento U-5-3H e indica que, embora o processo de sintese e translocacao possa ser menor, essas varias etapas podem se complementar dentro de cinco horas.
