AbstractAbstract
[en] The kinetics of heparin in normal Wistar rats using the radioactive tracer 51Cr, has been studied. The labeled and purified 51Cr-heparin was injected into rats intravenously and by intraperitoneal injection. In measuring the radioactivity of organs it was possible to conclude that the tissues rich in mast cells, liver and spleen, were found to take up the greater amounts of heparin. The curve that represents the logarithm of the concentration of heparin versus time is biexponential. The half-lives of the two exponential were determined. The volume of distribution, the rate constant and the renal clearance were determined by the values of the plasma levels and urinary excretions. The biological half-time, the turnover rate and the turnover time were determined by measuring the residual radioactivity of the total body and urinary excretions. With the data obtained from the mentioned experiments a compartmental model was performed in which the plasma is the central compartment for the distribution of the drug, exchanging with another extraplasmatic compartment and finally the drug being stored in reticulo endothelial system cells. Estudou-se a cinetica da heparina marcada com 51Cr. Injetou-se em ratos, pelas vias intravenosa e intraperitonial, heparina marcada com 51Cr e purificada. Medindo-se a radioatividade dos orgaos foi possivel concluir que os tecidos ricos em mastocitos, o figado e o baco captavam grandes quantidades de heparina. A curva que representa o logaritmo da concentracao da heparina versus tempo e biexponencial. Determinaram-se as meias-vidas das duas exponenciais. Determinaram-se o volume de distribuicao, a taxa constante e a depuracao renal pelos valores dos niveis plasmaticos e das excrecoes urinarias. Determinaram-se a meia-vida biologica, a taxa de renovacao e o tempo de renovacao atraves da medida da radioatividade residual de corpo total e pelas excrecoes urinarias. A partir dos dados obtidos, fez-se um modelo compartimental no qual o plasma e o compartimento central de distribuicao da droga, trocando com outro compartimento extraplasmatico e finalmente sendo estocada nas celulas do sistema reticulo-endotelial.
