AbstractAbstract
[en] Complete text of publication follows. The paper summarized the results on the ionizing radiation effects on some dehydrated enzymes of food interest as amylase, protease and pectinases. Irradiation has been performed to a linac (We ∼6 MeV) and to a 60Co γ-ray source, in the dose range of 0-50 kGy, to dose rates of ∼1.5 kGy/min for EB and ∼3.0 kGy/h for γ-ray irradiation. The radiation induced inactivation of enzymes and the modification of their kinetical properties has investigated. The enzyme activity (Ae), the Michaelis-Menten constant (KM) and the reaction velocity are influenced by dose in a specific manner presented by the obtained experimental dose-effect curves. The physical radio-induced modifications are investigated by spectrophotometry, EPR and neutron scattering. All EPR measurements were performed at room temperature by using an X band JEOL JES ME 3X spectrometer. The EPR spectra for irradiated and non-irradiated molecules, including the values of effective gyromagnetic factors and linewidths, are presented. The differences between EPR spectra give information about the mechanism of energy deposition by irradiation on proteins. Using a time-of-flight spectrometer (TOF base 210 cm) and a neutron flow of 2- 5 x 108 n/cm2 x s; λn ∼ 4 A; θScatt 0 deg - 80 deg; the neutron scattering spectra have been obtained. The spectra pointed out a very clear differentiation between the irradiated molecules and the non-irradiated ones, which should be assigned and described the irradiation effects on the hydrogen bound behavior. The spectra give also information on the molecule parameters (atomic distances, chemical bonds, diffusive motion)
