AbstractAbstract
[en] An increasing number of studies has shown that pre-treatment of seeds with non-thermal plasma (NTP) improves germination and seedling growth. However, very little is known about potential genotoxic effects of NTP in plants. We decided to study DNA damage in soybean seedlings (Glycine max L., var. Nizina) after NTP treatment of seeds with the use of comet assay (single cell gel electrophoresis). We found out that alkaline variant of comet assay was not suitable in our case because of very high DNA damage in negative control (non-treated seeds). Neutral variant, which detects smaller range of DNA damage than alkaline variant, was much more suitable. Using the optimized protocol, we discovered that NTP generated in ambient air did not induce significant DNA damage in soybean seedlings in any of the exposition times used. Nevertheless, it is necessary to perform additional experiments to further test our hypothesis. (authors)
